Clinical Utility

Anaerobic cultures are used in the diagnosis of infections caused by both aerobic and anaerobic bacteria. Clinical significance of bacteria isolated are evaluated based on specimen source, the organism's pathogenicity, the potential for skin contamination or colonization, and the number of other organisms isolated.

Specimen and Container Info

Specimen

Collection Container

Aspirate, Body Fluid, Exudate

Sterile Container or Clear Top Tube, No Additive

Tissue

Sterile Container

Wound

Eswabs or Alternate Anaerobic Transport

Note: Swabs are not preferred when tissue or aspirate can be obtained.

Collection Instructions
  • Collect specimen from prepared site using sterile technique. Avoid contamination with normal flora.
  • Pus or body fluid collected by syringe placed directly into Red Top, No Additive after decontaminating the top with alcohol.
  • Do not put fluid collected by syringe into Eswab.
  • Place tissue in sterile container covered with a small amount of sterile non-bacterial static saline.
Transport Instructions

Transport to the laboratory at room temperature.

Stability Requirements

Room Temperature

24 hours

Refrigerated

48 hours*

*For optimal recovery, specimens should be received by the laboratory within 24 hours.

Frozen

Unacceptable
Causes for Rejection
  • Specimens submitted in swabs not suitable for the recovery of anaerobes can only be accepted for aerobic culture.
  • Specimens collected from site with normal anaerobic flora will not be routinely accepted for culture. This includes Respiratory, Vaginal, Stool, Throat, and Urine Specimens.
Methodology
Culture with reflex to additional methods for identification and susceptibility if warranted
Limitations

Bacterial culture is not sufficient for the recovery of fungal organisms (except yeast) or Mycobacteria since extended incubation and special media are necessary for isolation.

Performing Location
Roble Road Core Laboratory

  • Bacterial Culture
  • Aerobe and Anaerobe Culture
