Clinical Utility

This assay is used to monitor Asparaginase therapy for sufficient enzyme activity, to assess silent inactivation by patient sensitization (activity assay) and to assess if a patient is producing antibodies against asparaginase (antibody assay).

Specimen and Container Info

Specimen Type: Serum

Collection Container: Gold Top, Serum Separator

Alternate Collection Container: Red Top, Plain

Desired Specimen Volume: 2 mL

Collection Instructions
  1. Collect a gold top, serum separator tube via venipuncture.

Processing and Submission Instructions

A completed ASPARAGINASE REQUISITION FORM is required for testing.

  1. Separate serum from cells within 2 hours of collection.
  2. Transfer 2 mL of serum to a clear aliquot tube (screw cap) and freeze.
  3. Indicate date and time of last injection on the submitted requisition.

Limitations

Measurement of samples are made against a standard curve generated from E. coli L‐asparaginase with each run. Calculations for Erwinaze®, Oncaspar®, Asparlas, and Rylaze® were empirically derived and utilize a correction factor that has been correlated to the E. coli L‐asparaginase activity.

The antibody assay reports the presence or absence of antibodies to asparaginase in the sample, but it does not distinguish between general antibodies towards asparaginase and neutralizing antibodies.
