Jackson-Weiss syndrome (JWS) - The craniosynostosis syndromes are a group of disorders sharing the premature fusion of one or more sutures of the skull. Often additional anomalies are associated. There are eight craniosynostosis disorders caused by mutations in three fibroblast growth factor receptor genes: FGFR1, FGFR2 and FGFR3. All are autosomal dominant. Penetrance varies with the specific craniosynostosis type. Many FGFR mutations are associated with advanced paternal age. Jackson-Weiss syndrome (JWS; MIM123150) is characterized by craniosynostosis, midface hypoplasia, mandibular prognathism, foot abnormalities with broad and medially deviated great toes and variable syndactyly, but no thumb findings. There is great clinical variability, and some affected individuals may not have craniofacial findings. Intelligence is normal. Jackson-Weiss syndrome is caused by FGFR2 mutations. Penetrance is high or complete. Prevelance is unknown. In some individuals identical FGFR2 mutations have been described in Jackson-Weiss, Crouzon and Pfeiffer syndromes. |
