Clinical Utility

Microsatellite Instability (MSI) PCR Analysis is performed on template DNA extracted separately from areas of normal and tumor tissue microdissected from formalin-fixed paraffin-embedded (FFPE) tissue under microscopic control. PCR amplification is performed for the microsatellite loci BAT25, BAT26, D2S123, D5S346, and D17S250, as recommended by the revised guidelines issued by the International Collaborative Group on HNPCC and the National Cancer Institute (Dietmaier et al., Cancer Res 1997, 57:4749-56; Boland et al., Cancer Res 1998, 58:5248-57; Uman et al., JNCI 2004, 96:261-68).

Specimen and Container Info
Specimen Type: FFPE Block, unstained slides with matched H&E slide
Processing and Submission Instructions
Formalin-fixed and paraffin-embedded tissue: Slides: 7-10 sections of 10 μm unstained and 1 matched H&E slide. Please take adequate precautions to prevent cross-contamination with other specimens during slide cutting and transport. If it is not possible to send slides, the tumor block is acceptable, although not preferred. Multiple different tests can be performed on the same specimen.
Transport Instructions
Transport to the laboratory at room temperature.
Stability Requirements

Room Temperature

FFPE Block, 5 years
Causes for Rejection
Specimens with less than 10% tumor content will be rejected.Do not send decalcified specimens, cytology smears or samples from hematologic malignancies.
Methodology
Next Gen Sequencing (NGS) | Multiplex Polymerase Chain Reaction (PCR)
