| Description | ||||||||||||||||||||||||||||||
|
Norrie disease (ND; MIM 310600) is a severe, X-linked recessive neurodevelopmental disorder caused by mutations in the NDP gene. It is characterized by early-onset blindness due to bilateral retinal dysplasia. Other eye findings include retinal folds, retinal detachment, retrolental masses commonly referred to as pseudoglioma, phthisical globe, microphtalmia and cataracts. Sensorineural hearing loss with onset in the second decade occurs in 25-30% of patients. Variable and progressive intellectual disability has been reported in about 50% of patients. Heterozygous females are usually unaffected, although a small number of females with retinal findings, vision loss, and mild sensorineural hearing loss have been reported. Norrie disease is allelic with one form of familial exudative vitreoretinopathy (EVR2; MIM 305390), a genetically heterogeneous disorder caused by deficiency in vascular development of the peripheral retina. |
