| Description | ||||||||||||||||||||||||||||||
|
Spondylo-megaepiphyseal-metaphyseal dysplasia (SMMD; MIM 613330) is a rare autosomal recessive skeletal dysplasia characterized by disproportionate short stature with a short and stiff neck and trunk. Patients with SMMD have relatively long limbs, large head with a broad face and widely spaced eyes. Some patients have flexion contractures of the distal joints. The main radiological findings include delayed and reduced ossification of the vertebral bodies, megaepiphyses and wide growth plates in the long tubular bones, and multiple pseudoepiphyses of the hands and feet. SMMD is caused by loss function mutations in NKX3-2 gene. It belongs to the NK3 homeobox family and plays a role in endochondral ossification of both the axial and appendicular skeleton. Achondrogenesis type 2 (ACG2; MIM200610), caused by mutations in COL2A1, is a major prenatal differential diagnosis. |
